Thymosin Alpha-1 (Tα1) is a naturally occurring 28-amino acid peptide originally isolated from thymic tissue. It is a key component of Thymosin Fraction 5, a partially purified extract from the thymus gland that was first characterized in the 1970s.

Tα1 plays a central role in immune system regulation, primarily through its effects on T-cell maturation and differentiation. It acts on toll-like receptors (TLR2, TLR9) on dendritic cells and enhances the immune system’s ability to respond to infections and tumors.

A synthetic version of Tα1 has been approved in over 35 countries for the treatment of hepatitis B and C, and as an immune adjuvant. It has not received FDA approval, though it has received orphan drug designation.
